Cutting off the oxygen supply to serious diseases

Tuesday, January 31, 2012

A new family of proteins which regulate the human body's 'hypoxic response' to low levels of oxygen has been discovered by scientists at Barts Cancer Institute at Queen Mary, University of London and The University of Nottingham.

The discovery has been published in the international journal Nature Cell Biology. It marks a significant step towards understanding the complex processes involved in the hypoxic response which, when it malfunctions, can cause and affect the progress of many types of serious disease, including cancer.

The researchers have uncovered a previously unknown level of hypoxic regulation at a molecular level in human cells which could provide a novel pathway for the development of new drug therapeutics to fight disease. The cutting-edge work was funded by the Biotechnology and Biological Sciences Research Council (BBSRC).

Proteins are biochemical compounds which carry out specific duties within the living cell. Every cell in our body has the ability to recognise and respond to changes in the availability of oxygen. The best example of this is when we climb to high altitudes where the air contains less oxygen. The cells recognise the decrease in oxygen via the bloodstream and are able to react, using the 'hypoxic response', to produce a protein called EPO. This protein in turn stimulates the body to produce more red blood cells to absorb as much of the reduced levels of oxygen as possible.

This response is essential for a normal healthy physiology but when the hypoxic response in cells malfunctions, diseases like cancer can develop and spread. Cancer cells have a faulty hypoxic response which means that as the cells multiply they highjack the response to create their own rogue blood supply. In this way the cells can form large tumours. The new blood supply also helps the cancer cells spread to other parts of the body, called 'metastasis', which is how ultimately cancer kills patients.

The scientists have identified a new family of hypoxic regulator proteins called 'LIM domain containing proteins' which function as molecular scaffolds or 'adapters' bringing together or bridging two key enzymes in the hypoxic response pathway, namely PHD2 and VHL. Both of these are involved in down-regulating the master regulator protein called Hypoxia-inducible factors (HIF1). The research has shown that loss of LIMD1 breaks down the bridge it creates between PHD2 and VHL and this then enables the master regulator to function out of control and thus contribute to cancer formation.

Molecular Oncologist, Dr Tyson Sharp, who carried out research for the project at The University of Nottingham's School of Biomedical Sciences, said: "The results from this research represent a significant advancement in our understanding of precisely how the hypoxic response works. It will help researchers develop better drugs to fight cancer and also other human diseases that are caused by low levels of oxygen within our body such as anaemia, myocardial infarction (heart attack), stroke and peripheral arterial disease. Further work in this fascinating area is now continuing at Barts Cancer Institute at Queen Mary University of London and will form the basis of a whole new additional research theme for my group."

Gingrich stays on attack in Florida with one day left (Reuters)

JACKSONVILLE, Florida (Reuters) ? U.S. Republican Newt Gingrich scrambled to revive his fading hopes in Florida on Monday, lashing out at rival Mitt Romney as a liberal party insider the day before the state's crucial presidential primary.

Polls showed Romney with a commanding lead in the state, where Gingrich's attacks on the former head of a private equity firm as an elite friend of Wall Street have failed to make an impression with Republicans who cast ballots on Tuesday.

Gingrich kept up the offensive on the campaign trail and in television interviews on Monday, pledging to stay in the White House race for the long haul and insisting Republicans eventually would rally to his candidacy.

"On big philosophical issues, he is for all practical purposes a liberal and I am a conservative and that's what this fight is going to be about all the way to the convention," he said of Romney on "CBS This Morning."

A win in Florida would be a vital boost for Romney, who was backed last week by several prominent conservatives and party leaders worried a Gingrich nomination would doom Republicans in the general election against President Barack Obama.

Gingrich said heavy spending by Romney, the former Massachusetts governor, and the Super PAC that supports Romney had killed the momentum Gingrich built with a double-digit win over Romney on January 21 in South Carolina.

"He can bury me for a very short amount of time with four or five or six times as much money, most of it raised in Wall Street from the guys who got bailouts from the government," Gingrich said on ABC's "Good Morning America."

A Quinnipiac University poll of likely Florida voters released on Monday indicated Romney has a 14-point advantage over Gingrich with broad-based support from across the Republican coalition including self-described conservatives, white evangelical Christians and Tea Party supporters.

Other polls also have shown Romney opening up a double-digit lead on Gingrich, the former speaker of the U.S. House of Representatives.

"If this margin holds up tomorrow, it's hard to see where Gingrich goes from here," Quinnipiac pollster Peter Brown said.

But the Gingrich campaign worked to spread the idea he was staying in the race long-term. "This race is just getting started," Martin Baker, Gingrich's national political director, said in a memo to reporters.

At least 1,114 delegates are needed to secure the Republican nomination in August. Florida is the largest state to hold a presidential primary so far this year, and 50 delegates are at stake in a winner-take-all format that will decide who faces Democratic President Barack Obama in November.

'LONG WAY TO GO'

But Baker said even a Romney win in Florida will give him just 7 percent of the delegates needed to claim the nomination. The next contest after Florida will be Nevada on Saturday, followed by Minnesota, Colorado and Missouri on February 7.

"There is a long way to go before either candidate clinches the nomination, and this campaign will continue for months," he said. "The campaign is shifting to a new phase where opportunities are not limited to a single state."

Talking to reporters on Monday, Romney noted Gingrich's growing interest in the campaign's long haul.

"That's usually an indication that you think you're gonna lose. When you say 'I'm gonna go on no matter what happens,' that's usually not a good sign," Romney said.

Gingrich said opposition to his candidacy by party insiders was a badge of honor and a sign of how threatening his candidacy was the elite.

"They recognize I'm a genuine outsider. I know a lot about Washington having served as speaker but have none of the establishment ties and I will shake the system up. They don't want to be shaken up," he said on CBS.

Both candidates crisscrossed Florida in a final hunt for votes on Monday. The two other remaining Republican contenders, former U.S. Senator Rick Santorum and U.S. Representative Ron Paul, are looking beyond Florida to the next races in Minnesota, Colorado and Nevada.

After bickering with Romney over which of them was best suited to extend former President Ronald Reagan's legacy of conservatism, Gingrich brought Reagan's son, Michael, with him on the campaign trail on Monday to bolster his case.

Gingrich said Reagan's presence should end questions from the party establishment about his links to Reagan.

"I figured if his son was prepared to campaign with me, for anybody with an open mind, that should settle that issue once and for all," Gingrich said in Jacksonville.

Reagan said he owed Gingrich a campaign appearance "because of what he's done for the Republican Party over the years. I remember him back in the 1980s during the Reagan revolution."

7 steps for business success with big data ? Cloud Computing News

No longer the new technology on the block, big data continues to generate significant buzz. ?Technologies such as Hadoop and HBase are seeing rapid growth, analysts are experimenting with new techniques and approaches, and business leaders are adapting their business models to rely more on the power of big data. McKinsey calls big data the ?next frontier? for business, with the potential to transform business in the same way the Internet did over the past 15 years.

To take advantage of that potential, business leaders need to know what steps to take in order to make maximum use of their data asset. Business success with big data is not just about choosing the right cloud technologies or hiring smart data scientists ? it?s about creating a business-centric approach that connects a company?s data to its business strategy, enables continual improvement, and follows through to impact processes, margins, and customer satisfaction.

In my experience with big data I have developed seven steps that can help any business leader drive more success with their big data initiatives.

1. Create a strategy for your data

Your data needs a strong strategy, one that connects to and underpins your business strategy and also integrates with department-level accountabilities. Develop a plan for where you want to be at each milestone, define your future capabilities clearly, and describe how your data capabilities will be utilized. Then hold your users accountable to where and how they will use the new capabilities, and what business impact they will drive.

For example, if you can use big data to improve in-store sales, you need to not only work with store managers to define capabilities that connect to their strategy, but also ensure the managers are held accountable to using the data capabilities correctly and delivering the intended impact. Doing so connects both your and their goals to the overall business strategy, helps create more usable capabilities, and ensures any needed iterations will be done jointly. A well-conceived data strategy will give you the most bang for the buck on your data investment.

2. Design for agility

Big data systems are just that?big, which means they tend to be inflexible. A great BI system, by definition, will cause a business to change, which in turn will require the BI system to change. Thus, your systems need to adapt quickly to keep pace with your business. Twelve- or 18-month release cycles are appropriate for certain parts of your system, but 3- or 6-month cycles may be appropriate for others. Carefully analyze each component of your big data system, and design for the right amount of agility you need.

You may decide to build higher levels of automation into the layers of your stack that change slowly, while reserving configuration-based approaches for layers of your stack that need to change quickly. In general, the top layers of your stack (e.g., user interfaces and reporting tools) need to be more agile than the bottom layers of your stack (e.g., data collection and storage), but many exceptions to this rule exist. Only careful analysis and understanding of your current and future uses of data will enable you to make the right decisions on agility. Designing for agility will enable your big data investment to keep pace with, and even lead, your business.

3. Understand latencies

Latency is a challenge in traditional BI systems, and big data only amplifies the problem. Big data solutions tend to be architected first as batch systems, with lower latency capabilities being addressed afterwards. Don?t save latency for last ? analyze your key use scenarios in terms of latencies, and connect them clearly to business drivers. Focus on delivering the right latency for each need, including the value being driven, and let those needs drive your design. ?Certain low latency needs may require bypassing your big data system temporarily, sharing directly between systems in order to deliver specific scenarios.

For example, if your customers tend to interact with system A and system B in parallel or in quick sequence, these two systems may need to share data directly. The data can then be written into the big data system in time to be used by other systems. Delivering data on a real-time or near-real-time basis can be very expensive; thus, it?s better to think in terms of ?right-time? data targeted to each need.

Describe latency requirements in detail, and ensure the business justification is sound. Understanding latencies will enable you to deliver data exactly when it?s needed, while keeping costs under control.

4. Invest in data quality and metadata

Data quality in any system is a constant battle, and big data systems are no exception; however, big data systems require much more automation and advance planning. ?You should first ensure that data quality is not treated as a project or initiative, but as a foundational layer of your data stack that receives adequate resourcing and management attention. ?Second, build in multiple lines of defense ? from data mastering (where, for example, you are creating customer accounts) to data collection (where you are recording all of that customer?s interactions with you) to metadata (where you are organizing and dimensionalizing the data to aid in future reporting and analysis). ?Third, automate both the processes that identify and elevate data quality issues, and the measurement and reporting of data quality progress.? Empower your data quality team with tools that solve problems at high scale, such as diagnostic and workflow tools.? Efficient data quality practices will enable your big data system to earn its place as a trusted input for key business processes.

5. Get good at prototyping

The data sizes in most big data systems are too large to work with all at once, so it?s typically wiser to build small-scale prototypes to iron out the wrinkles and ensure you are meeting customer requirements. If you are building complex data integrations, online algorithms, or user interfaces, prototyping allows you to learn at a smaller and less costly scale. What?s more, prototypes can be shared early with your user base, which generates valuable feedback as well as excitement.

Prototyping requires somewhat unique skills that you will need to build and refine over time. Prototypers need to be able to move quickly, figure out new designs and technologies, understand user scenarios, actively solicit feedback, and not be afraid to fail. They need to be creative in their approach to solving problems, while still rooted in sound data mechanics. Why is prototyping better than wireframes or feature lists? Since prototypes are ?real,? your users will give you better feedback; at the same time you will also understand some of the challenges you will face as you build the full-scale version. Building a strong prototyping capability will help you increase innovation and speed, while reducing the cost of mistakes.

6. Get great at sampling

Sampling will save you a lot of time if you learn how to do it correctly. There are many use cases for which sampling is an effective alternative to using full census (100 percent) data. Certain needs such as creating personalized experiences for each customer, or calculating executive accountability metrics, are not appropriate for sampling. But for many other needs, sampling is a viable option.

For example, understanding product or feature performance, looking at patterns and trends over time, and filtering for unexpected anomalies can typically be done on sampled data. One approach is to collect 100 percent of the data, but do most of your analysis on samples, and then confirm important conclusions on the full data set. Once you establish process flows to pull sampled data into standard tools such as Excel and/or SQL, you will see analyst productivity increase substantially, which will save you time and money and increase the job satisfaction of your analysts.

To get great at sampling, you need to do three things: first, develop standard sampled data sets that help your analysts address large swaths of business questions, updating them regularly; second, make sure you have at least one highly qualified individual (i.e. a statistician) who can ensure the data is being sampled correctly and results are not misapplied; and third, educate decision makers on the benefits and limitations of sampling so they can get comfortable making decisions with sampled data. The effective use of sampling increases productivity while delivering equivalent business value.

7. Ask for regular feedback

Big data is a learning process, both in terms of managing the data and in driving business value from its contents. Your internal user base is a valuable source of feedback and integral to your learning and development process. Your prototyping program will be a source of feedback, but you should also survey your users and benchmark your progress over time. Areas such as usability, data quality, and data latency are all categories within which users will give you feedback. In addition, you should ask for ad hoc feedback from every level of your stakeholder organizations so they see your commitment to making their business better.

As your data asset?s reputation grows, your stakeholders will give you more and better feedback, which will allow you to develop integrated goals and roadmaps, and drive more business benefit as a result. Regular feedback ensures your big data system is tightly integrated into business decision making, so it can play a lead role in business improvement.

Following the above steps will help you build more effective big data capabilities, saving you time and money, and driving maximum ROI for your business. The big data frontier is here; breaking through it requires an understanding of which steps will help you drive the most impact.

British police arrest 5 in tabloid bribery probe

(AP) ? British police searched the offices of Rupert Murdoch's British newspapers Saturday after arresting a police officer and four current and former staff of his tabloid The Sun as part of an investigation into police bribery by journalists.

The arrests spread the scandal over tabloid wrongdoing ? which has already caused the closure of one tabloid, the News of the World ? to a second Murdoch newspaper.

London's Metropolitan Police said two men aged 48 and one aged 56 were arrested on suspicion of corruption early in the morning at homes in and around London. A 42-year-old man was detained later at a London police station.

Murdoch's News Corp. confirmed that all four were current or former Sun employees.

A fifth man, a 29-year-old police officer, was arrested at the London station where he works.

The investigation into whether reporters illegally paid police for information is running parallel to a police inquiry into phone hacking by Murdoch's now-defunct News of the World.

Officers were searching the men's homes and the east London headquarters of the media mogul's British newspapers for evidence.

Police said Saturday's arrests were made as a result of information provided by the Management and Standards Committee of Murdoch's News Corp.

News Corp. said it was cooperating with police.

"News Corporation made a commitment last summer that unacceptable news gathering practices by individuals in the past would not be repeated," it said in a statement.

A dozen people have now been arrested in the bribery probe, though none has yet been charged.

They include former Rebekah Brooks, former chief executive of Murdoch's News International, ex-News of the World editor Andy Coulson ? who is also Prime Minister David Cameron's former communications chief ? and journalists from the News of the World and The Sun.

Two of the London police force's top officers resigned in the wake of the revelation last July that the News of the World had eavesdropped on the cell phone voicemail messages of celebrities, athletes, politicians and even an abducted teenager in its quest for stories.

Murdoch shut down the 168-year-old tabloid, and the scandal has triggered a continuing public inquiry into media ethics and the relationship between the press, police and politicians.

An earlier police investigation failed to find evidence hacking went beyond one reporter and a private investigator, but News Corp. has now acknowledged it was much more widespread.

Last week the company agreed to pay damages to 37 hacking victims, including actor Jude Law, soccer star Ashley Cole and British politician John Prescott.

>> thanks be p.

>> i went on your website just to read some of the comments that people had regarding the story. it was a mixed bag. it was a lot of concern about some of the details which which you the alleged victim was never identified. it was no formal complaint filed. what is some of the reaction you're getting be.

>> the point of his statement doesn't really contradict what's in our article except for one item. he's saying at the time he made his statement withdrawing his candidacy that he was still active. our source is saying no. that's really the only contra contradiction between what he is saying.

>> one of the comments says it's raises a lot of concern. publishing such a stoirp in absence of name, sources or formal charges much less convictions here. this is the concern that you're talking about this young man's life, career. he's released a statement. you have sources but none have been identified including this alleged victim here.

>> it's a legitimate concern. obviously, it's a decision that we weighed very carefully. i think had he not been a rhodes candidate, this isn't something we reported on. the impression was left out there. i'm not saying that he necessarily lied about it. the impression was left out there and reported in the national media over and over again that he had said, i'm not going to do the interview, so that i can play in the harvard game.

>> in your report, he didn't leave the impression. that was a conclusion made by reporters who covered the story. he, i believe to your point, did not say. he didn't lie. he didn't deceive here.

>> right. his statement was pretty carefully worded. it said i'm going to play in the harvard game. i've withdrawn my application. it didn't say i'm playing because i with draw my application.

>> let me read quickly yale 's response. yale says all schools are prohibited by federal law from disclosing the names of students involving in an allegation of misconduct. the applications by students for scholarships and fellowships is confidential. yale has a policy in place. rhodes trust has no comment. new haven police didn't, no formal complaint has been filed. i'll bring it back again, was the times fair to this young man here?

>> i think so. i think that he's a public figure out there in the public eye and was a misimpression created about him. again, i think were we not a candidate, we wouldn't have done a story like there.

>> back to the woman who made a complaint with the university wide committee on sexual misconduct . this happened back in july. again, with their not being a criminal charge here, was there concern by you and the staff who put this story together that with no formal complaint here, this may be perceived by people not -- who don't care about football or yale , that this young man was not being treated appropriately in the reporting of this story?

>> it was something we considered. we decided it was worth going forward. we make no judgment as to the validity of the allegation. it was made through an informal process that doesn't include any disciplinary penalty, any finding of guilt. that's the nature of that process.

>> thank you very much. i appreciate you coming onto talk about this story. people have beenfired up about this. i think it's great you came onto talk about the reporting here. thank you.

>> thank you.

US women's soccer team qualifies for Olympics (AP)

VANCOUVER, British Columbia ? In the end, what matters is that the U.S. women's soccer team is going to the Olympics. What the players won't soon forget is how hard they had to work get there.

The Americans booked their way to London on Friday night with a 3-0 victory over Costa Rica in the semifinals of the CONCACAF qualifying tournament, a game more suspenseful than most anyone expected.

Tobin Heath scored in the 16th minute to give U.S. all the scoring it needed, and goals by Carli Lloyd (72nd) and Alex Morgan (89th) put the game away late.

But the top-ranked Americans were less crisp than when they were beating teams by a combined 31-0 earlier in the tournament and drawing criticism for running up the score.

Costa Rica is ranked No. 41 in the world, has never qualified for an Olympics or a World Cup and has never scored on the U.S. in eight meetings. Las Ticas proved to be scrappy opponents, however, occasionally frustrating the Americans with physical play and just missing on two solid scoring chances in the first half in the London-or-bust match. As the possibility of an upset lingered deep into the second half, the underdogs gained the rousing support of the Canadian fans at BC Place.

In the end, a speedy, unflappable quartet of American defenders ? Kelley O'Hara, Rachel Buehler, Christie Rampone and Amy LePeilbet ? combined with goalkeeper Hope Solo to keep Costa Rica out of the net. Solo played despite a slightly pulled right quadriceps that had been bothering her all week.

The U.S. will be the two-time defending champions in London, having taken gold in Athens in 2004 and in Beijing in 2008. It will be the third straight Olympics in which the Americans will be trying to make amends for World Cup disappointment from the previous year. They finished second at last year's World Cup in Germany, losing to Japan in the final.

The victory also puts the Americans into the tournament final Sunday, a bragging-rights-only game against the winner of Friday's late game between Mexico and Canada.

The Americans had scored so easily in the tournament that it seemed odd to see the game scoreless until the 16th minute, when a set piece produced the first goal. Lauren Cheney's corner kick was headed down at the far post by Shannon Boxx. Goalkeeper Erika Miranda made the save but deflected the ball to Heath, whose looping header was her fifth career U.S. national team goal.

Costa Rica, outscored 34-0 in the seven previous games against the U.S., nearly tied the game after a giveaway by Buehler set up Fernanda Barrantes with a clean look from 15 yards in the 20th minute, forcing Solo to the ground to make the save.

Then, in the 27th, Carol Sanchez launched a 30-yarder that clanged off the frame at the intersection of the post and the crossbar. With Solo on the ground, Buehler fought off Barrantes to keep the striker from getting the rebound with a clean shot at an open net.

Costa Rica finally had its hopes deflated in the 72nd, when Abby Wambach's chip shot was cleared off the line by Daniela Cruz and out to Lloyd, whose left-footer from the top of the 18-yard box doubled the lead.

Morgan, a second-half substitute, chipped in the insurance goal shortly before the final whistle.

Even with the closer-than-expected result, the Americans have evoked the good old days at this tournament with their mostly lopsided scores. While that's hardly surprising given the slow development of women's soccer in parts of North and Central American and the Caribbean, it's also indicative the U.S. still have the deepest, most talented team in the world.

But Pia Sundhage's team arrived in Canada with a bit of apprehension. The Americans, having become somewhat complacent from years of uncontested success in the region, were stunned in a World Cup qualifier by host Mexico in November 2010, forcing them into a home-and-away playoff with Italy just to get for the World Cup. Also, the format for Olympic qualifying is such that everything hinges on one game ? the do-or-die semifinals ? regardless of how a team performs in the rest of the tournament.

Determined to take nothing for granted, the Americans have been full throttle for every game. They set a U.S. team record for goals in a game in a 14-0 win over the Dominican Republic, then nearly matched the feat in a 13-0 rout of Guatemala. Then came a 4-0 win a much anticipated rematch with Mexico to set up the semifinal against Costa Rica.

(AP) ? Nike founder and chairman Phil Knight got a standing ovation at Joe Paterno's public memorial Thursday for defending the late coach's response to an accusation of child sex abuse against a former assistant.

At a ceremony filled with lavish praise that would likely have embarrassed the school's beloved coach, Knight waded into a subject that had been virtually ignored since Paterno died on Sunday from lung cancer ? the immediate cause of his firing after 46 seasons leading Penn State's football team to a record-setting 409 wins.

University trustees have said a major reason Paterno was ousted Nov. 9 was that he failed to alert anyone beyond his two superiors at the school when he was told of an abuse allegation against former assistant Jerry Sandusky in 2002. Sandusky was charged with dozens of child sex abuse counts on Nov. 5, two school officials were charged with perjury and former school president Graham Spanier was pushed out the same night as Paterno.

Commenting on those events, Knight said, "it turns out (Paterno) gave full disclosure to his superiors, information that went up the chains to the head of the campus police and the president of the school. The matter was in the hands of a world-class university, and by a president with an outstanding national reputation."

Knight added, "...this much is clear to me. If there is a villain in this tragedy, it lies in that investigation and not in Joe Paterno."

The remark drew a standing ovation from the crowd of 12,000 at the Bryce Jordan Center ? with Paterno's widow, Sue, and former defensive coordinator, Tom Bradley, among those rising to their feet.

"Who is the real trustee at Penn State University?" Knight asked.

The campus has been torn by anger over the Sandusky scandal and Paterno's firing, but this week thousands of alumni, fans, students and former players in Happy Valley have remembered Paterno for his stellar career, his love for the school and his generosity.

That made Knight's remarks even more surprising.

Paterno's son and former quarterbacks coach, Jay, also got a standing ovation near the end of the ceremony when he imitated his raised-in-Brooklyn father's voice, telling the audience to "Sit down! Sit down!"

Then, growing serious, he told the crowd, "Joe Paterno left this world with a clear conscience."

Earlier in ceremony, the focus was praise for Paterno and fond memories.

"Bless us this day as we honor and celebrate one of your greatest gifts to the world ? Joe Paterno," the Rev. Matthew Laffey said in the opening prayer.

A short time later, after a video montage, former star Penn State quarterback Todd Blackledge said, "No one individual has ever done more for a university anywhere in the country than what Joe Paterno did for this school."

The line drew applause, and Blackledge was followed on the podium in the darkened arena by Lauren Perrotti, a Penn State student and Paterno fellow.

Charles V. Pittman, speaking for players from the 1960s, called Paterno a lifelong influence and inspiration.

"Now, with grown children grandkids and 42 years removed from my playing days, I thought Joe Paterno had taught me all that he could teach me. I was wrong," he said. "Despite being pushed away from his beloved game, and under the extreme pressure of the events of the past few months, Joe's grace was startling."

Pittman said Paterno pushed his young players hard, once bringing Pittman to tears in his sophomore year. He realized later that the coach was not trying to break his spirit but instead was "bit by bit building a habit of excellence."

"He was building a proud program for the school, the state and the hundreds of young men he watched over for a half century," said Pittman, senior vice president for publishing at Schurz Communications Inc., an Indiana-based company that owns television and radio stations and newspapers, and a member of the Board of Directors of The Associated Press.

Thursday's event brought to a close the public mourning period for Paterno.

Public viewings were held Tuesday and Wednesday morning at a campus spiritual center, followed by a funeral Mass, procession and burial for Paterno that afternoon.

Comodo Antivirus 2012

Going online without an antivirus utility to guard your back is just foolish. Even the safest, biggest site can be hacked so that just visiting it earns you an unwanted malware attack. Cost is no excuse, not when there are free choices like Comodo Antivirus 2012. But be warned: Comodo's behavior-based Defense+ module will enlist you as an essential member of the malware-fighting team.

Installation
Comodo Antivirus installed without difficulty on all but one of my 12 malware-infested test systems. The installation process felt rather long, with a required reboot followed by a lengthy antivirus signature update. Immediately on finishing the update, Comodo launched a full scan. That makes sense?the first thing most users want to do is make sure there are no lurking threats.

On the one problem system, Comodo's installation failed with an error message every time due to interference by malware. Installing in Safe Mode didn't work, so I cleaned up the system using Comodo Cleaning Essentials (free, 4.5 stars). After that, the antivirus installed without trouble.

Average Cleanup
At the end of each malware cleanup session, Comodo reported that it had found malware and offered to have a GeekBuddy expert handle the cleanup process. I found this a little misleading, since GeekBuddy service is not free. In each case, I clicked away the offer.

Comodo lists all the malware threats it found, along with the associated file and Registry traces. It assigns each found item a risk level, but marks threats of all levels for removal. In a few cases, it needed a reboot to complete the cleanup.

Comodo detected 85 percent of the threats, just a hair below average. Poor removal of the traces it did find earned it a mediocre malware removal score, 5.4 of 10 possible points. It left behind executable traces for close to half the detected threats, and for nearly half of those at least one process was still running after supposed removal. Comodo left behind all the non-executable traces for many of the rest. AVG Anti-Virus Free 2012 (free, 4 stars), our Editors' Choice for free antivirus, scored 6.5 in this test.

This antivirus proved especially weak against rootkits, detecting 85 percent of them and scoring a low, low 3.9 points, the same as McAfee AntiVirus Plus 2012 ($39.99 direct, 3.5 stars). Comodo Cleaning Essentials removed two rootkits in order to allow installation of Comodo Antivirus. Of those detected by the antivirus alone, all remained running with their rootkit technology still active.

Comodo Antivirus did detect all the scareware samples, but most products manage that feat. Comodo's score of 8.3 for scareware removal is actually low, given that well over half the current competition scored 9.5 or higher. Norton AntiVirus 2012 ($39.99 direct, 4.5 stars) and Malwarebytes' Anti-Malware Free 1.51 (free, 4 stars) aced this test with a perfect 10.

For an explanation of my testing technique, see How We Test Malware Removal.

Music has a greater presence at Sundance festival (AP)

PARK CITY, Utah ? The 2012 Sundance Film Festival in Utah isn't just about movies. Festival director John Cooper says organizers added more music to the 11-day festival this year to encourage relationships between independent filmmakers and musicians.

The ASCAP Music Cafe on Park City's Main Street hosts artists every day during the festival and also nightly concerts. And parties around town feature established and emerging musicians, such as R&B singer Estelle and Swedish DJ Sebastian Ingrosso.

Documentaries about Neil Young, Paul Simon and Ice-T are also part of the festival lineup.

ASCAP Music Cafe producer Loretta Munoz says the festival's mission is similar to that of the songwriters' organization she represents ? to foster great art by encouraging and supporting independent artists.

The Sundance festival continues through Sunday.

Armie Hammer arrested for pot possession

Authorities say Hollywood actor Armie Hammer was arrested at a border patrol checkpoint in Texas after a drug sniffing dog discovered marijuana in his car.

The 25-year-old, who starred with Leonardo DiCaprio in "J. Edgar," spent about a day in jail before paying a $1,000 bond.

Arrest records show he had 0.02 ounces of marijuana, three medicinal marijuana cookies and one brownie when arrested Nov. 30 in Sierra Blanca, a few miles from the Mexican border.

El Paso's district attorney declined to prosecute because a felony requires more than 4 ounces of actual marijuana. The county attorney could pursue lesser charges since the case is going back to the local sheriff, but Hammer's lawyer Kent Schaffer says no charges have been presented.

Hammer also starred in "The Social Network."

(AP) ? New York Mayor Michael Bloomberg says police used terrible judgment when they showed a film about Muslims during counterterrorism training seminars.

Bloomberg said Tuesday that police had stopped showing the movie, "The Third Jihad." It discusses extremists and their plans for establishing an Islamic regime.

Police documents obtained by a think tank at New York University say the movie was shown on a continuous loop while officers were signing in for counterterrorism training in late 2010. Nearly 1,500 officers may have seen the movie. Police originally said only a few officers had seen it.

Deputy Commissioner Paul Browne says that the movie was not approved for training courses and that the decision to play it was made by a sergeant. He said she had been reprimanded.

Kim K: Wedding was a 'bad business decision'

Three months after filing to end her 72-day marriage to Kris Humphries, Kim Kardashian is still forced to defend rumors that her televised nuptials -- and her union to the NBA star -- was a sham.

During a Monday cohosting gig with Kelly Ripa on "LIVE! With Kelly," the 31-year-old reality star mogul slammed rumors she tied the knot just for fame and fortune.

PHOTOS: Kris and Kim's doomed wedding

"Being a hopeless romantic, I wanted to believe in something so badly. If you think I really made all that money that everyone claimed we made on the wedding and that it was for TV...I'm a smart businesswoman, I would have stayed married longer!" Kardashian reasoned to Ripa during the morning show's Host Chat segment. "This was a bad business decision. I really didn't think following my heart would create this much backlash."

NEWS: Medium John Edward helps Kim realize she should end her marriage

Pulling the plug on her marriage so quickly was a necessity for the reality star. "I definitely tried everything I could [to save it], but also heard stories of people who had kids and stayed married for years and it didn't end up working out," she said. "I want babies and that forever love and if I felt in my heart that this wasn't right, why wait years for the same result?"

PHOTOS: Hollywood's shortest marriages

In the wake of her Oct. 31 divorce filing from Humphries, 26, Kardashian says she found herself leaning heavily on her mom, Kris Jenner, who divorced her first husband, Robert Kardashian, Sr., in 1990.

PHOTOS: LOL! Kelly Ripa and Nick Lachey reenact Kim's wedding day

"My mom and dad had four kids and they split up. I couldn't even fathom it at the time but as I got older, I began to understand her a bit more," Kardashian told Ripa. "Thinking of myself in that situation, if my mom had four kids and she was able to get out of a situation that she felt like didn't fit her, I think anybody could."

PHOTOS: The Kardashian family photo album

Still, the suddenly-single star hasn't given up on finding her happy ending.

"I just have to rewrite my fairytale," she told Ripa.

`Underworld' sinks teeth into box office with $25M (AP)

LOS ANGELES ? Kate Beckinsale is back with a vengeance, with her latest "Underworld" movie opening at No. 1 this weekend.

"Underworld Awakening" made an estimated $25.4, distributor Sony Screen Gems reported Sunday.

This is the fourth film in the vampire action saga. Beckinsale starred in the first two movies as the warrior Selene, then bowed out of part three but returned for this latest installment. "Underworld Awakening" was shown for the first time in 3-D as well as on IMAX screens, where it made $3.8 million. That's 15 percent of the film's weekend gross, which is a record for an IMAX digital-only run.

Sony had hoped the film would end up in the low-$20 million range. But Rory Bruer, the studio's president of worldwide distribution, says the fact that it did even better ? despite a snow storm that hit much of the Midwest and East Coast ? primarily has to do with Beckinsale's return.

"She is such a force. Her character ? you just can't take your eyes off of her. I know the character is very dear to her, as well, and she just kills it," Bruer said. "The 3-D aspect of the film also brings something, makes it a fun, visceral ride."

Opening in second place was "Red Tails" from executive producer George Lucas, about the Tuskegee Airmen who were the first black fighter pilots to serve in World War II. It made an estimated $19.1 million, according to 20th Century Fox, which was well above expectations; the studio had hoped to reach double digits, said Chris Aronson, executive vice president of domestic distribution.

"I believe what George Lucas has stated all along: This is an important story and a story that must be told. It is a true story of American heroism and valor and audiences have really responded to this message," Aronson said. "People want to feel good about themselves, they want to be uplifted. We have enough hard crud going on in this country right now. Times are tough, and if we look back and are told a story of some really fantastic deeds, that's really compelling moviegoing."

Hollywood.com analyst Paul Dergarabedian said a grass-roots effort to get groups of people into the theaters to see "Red Tails," along with positive word-of-mouth, helped its strong showing. The film saw an uptick from about $6 million on Friday to $8.65 on Saturday.

Overall box office is up 31 percent from the same weekend a year ago, Dergarabedian said, thanks to new releases as well as movies like "Extremely Loud & Incredibly Close," which had limited runs for awards consideration at the end of 2011 and are now expanding nationwide. The 9/11 drama from Warner Bros., starring Tom Hanks and Sandra Bullock, came in fourth place with $10.5 million.

Last week's No. 1 film, the Universal smuggling thriller "Contraband" starring Mark Wahlberg, dropped to the No. 3 spot with $12.2 million. It's now made $46.1 million in two weeks. Meanwhile, Steven Soderbergh's international action picture "Haywire" from Relativity Media, starring mixed martial arts superstar Gina Carano in her first film role, opened in fifth place with $9 million, which was above expectations.

"This is a great, perfect January weekend. You've got these holdover films and newcomers creating an overall marketplace that people are really responding to," Dergarabedian said. "It sounds clich? but this marketplace really has something for everyone."

As for worldwide box office, "The Twilight Saga: Breaking Dawn ? Part 1" has now crossed the $700 million mark. The first half of the finale of the girl-vampire-werewolf love triangle franchise has grossed an estimated $701.3 million in global box office receipts since its release last November, according to Lionsgate, which recently acquired Summit Entertainment, which distributes the series.

Estimated ticket sales for Friday through Sunday at U.S. and Canadian theaters, according to Hollywood.com. Where available, latest international numbers are also included. Final domestic figures will be released Monday.

1. "Underworld Awakening," $25.4 million ($13.4 million international).

2. "Red Tails," $19.1 million.

3. "Contraband," $12.2 million.

4. "Extremely Loud & Incredibly Close," $10.5 million.

5. "Haywire," $9 million.

6. "Beauty and the Beast (3-D)," $8.6 million.

7. "Joyful Noise," $6.1 million.

8. "Mission: Impossible ? Ghost Protocol," $5.5 million. ($9.4 million international).

9. "Sherlock Holmes: A Game of Shadows," $4.8 million. ($18.1 million international).

10. "The Girl With the Dragon Tattoo," $3.75 million ($15.7 international).
